Psychophysical thresholds for the detection of a 300-Hz burst of vibration applied to the thenar eminence were measured for stimuli applied to the skin through 1.5?cm2 and through 0.05?cm2 contactors. Thresholds were approximately 13?dB lower when the area of the contactor was 1.5?cm2 than when it was 0.05?cm2. The difference between the thresholds measured with the large and small contactors was significantly reduced when only the lowest thresholds obtained in the testing sessions were considered. This result supports the hypothesis that one component of spatial summation in the P channel is probability summation. In addition, threshold measurements within a session were less variable when measured with the 1.5?cm2 contactor. We conclude that spatial summation in the P channel is a joint function of two processes that occur as the areal extent of the stimulus increases: probability summation in which the probability of exceeding the psychophysical detection threshold increases as the number of receptors of varying sensitivities increases, and neural integration in which neural activity originating from separate receptors is combined within the central nervous system rendering the channel more sensitive to the stimulus.  相似文献

Many studies have examined the effects of spatial heterogeneity in light supply on clonal plants in terrestrial environments, but few have examined those in aquatic conditions. In a greenhouse experiment, we grew the rhizomatous submerged macrophyte Vallisneria spiralis L. in containers in three homogeneous light treatments (100%, 65%, and 30% of ambient light coded as high, medium, and low light, respectively) and two heterogeneous ones differing in patch scale (small and large patch). The growth of V. spiralis decreased significantly with decreasing light availability. In the low light conditions, V. spiralis allocated greater biomass to shoots and developed elongated leaves. In the patchy treatments, ramets distributed in the light‐rich patches had significant costs in the large patch treatment, but not in the small patch treatment, while both small and large patch treatments had no significant benefits in the light‐poor patches. We conclude that V. spiralis could escape from adverse habitats and occupy the favorable habitats. Providing the same amount of light, responses of V. spiralis to different patch scales were different at the patch level, but not at the whole clone level. Together, growth of V. spiralis could not benefit from different patch scales in heterogeneous environments at the patch as well as the whole clone level.  相似文献

Sp100是核颗粒ND10的组成蛋白,在哺乳动物细胞中广泛存在.Sp100参与多种细胞生理病理过程,如转录调控、细胞内抗病毒免疫等.利用酵母双杂交系统,我们发现了Sp100的互作蛋白HIV-1整合酶,免疫共沉淀实验进一步证实了Sp100与 HIV-1整合酶的互作,细胞内荧光共定位实验也证实了二者在细胞内部分共定位.此外,突变体实验表明,Sp100的C端300~480氨基酸和HIV-1的催化结构域是两个蛋白质的互作区域.利用siRNA降低细胞内Sp100的表达量,可以增加HIV-1整合酶介导的病毒的整合,反之,细胞内过表达Sp100则会降低HIV-1整合酶介导的病毒的整合.这是首次发现Sp100可以和HIV-1整合酶发生相互作用,并进而抑制病毒的整合.我们发现了Sp100作为HIV-1整合酶互作蛋白的新功能,并扩展了细胞防御病毒感染的相关研究.  相似文献

Sense of agency, the experience of controlling external events through one''s actions, stems from contiguity between action- and effect-related signals. Here we show that human observers link their action- and effect-related signals using a computational principle common to cross-modal sensory grouping. We first report that the detection of a delay between tactile and visual stimuli is enhanced when both stimuli are synchronized with separate auditory stimuli (experiment 1). This occurs because the synchronized auditory stimuli hinder the potential grouping between tactile and visual stimuli. We subsequently demonstrate an analogous effect on observers'' key press as an action and a sensory event. This change is associated with a modulation in sense of agency; namely, sense of agency, as evaluated by apparent compressions of action–effect intervals (intentional binding) or subjective causality ratings, is impaired when both participant''s action and its putative visual effect events are synchronized with auditory tones (experiments 2 and 3). Moreover, a similar role of action–effect grouping in determining sense of agency is demonstrated when the additional signal is presented in the modality identical to an effect event (experiment 4). These results are consistent with the view that sense of agency is the result of general processes of causal perception and that cross-modal grouping plays a central role in these processes.  相似文献

Integrative gene transfer is widely used for bioproduction, drug screening, and therapeutic applications but usual viral methods lead to random and multicopy insertions, contribute to unstable transgene expression and can disturb endogenous gene expression. Homologous targeting of an expression cassette using rare‐cutting endonucleases is a potential solution; however the number of studied loci remains limited. Furthermore, the behavior and performance of various types of gene cassettes following gene targeting is poorly defined. Here we have evaluated three loci for gene targeting, including one locus compatible with the proposed Safe Harbor criteria for human translational applications. Using optimized conditions for homologous gene targeting, reporter genes under the control of different promoters were efficiently inserted at each locus in both sense and antisense orientations. Sustainable expression was achieved at all three loci without detectable disturbance of flanking gene expression. However, the promoter, the integration locus and the cassette orientation have a strong impact on transgene expression. Finally, single targeted integrations exhibited greatly improved transgene expression stability versus multicopy or random integration. Taken together, our data suggest a potential set of loci for site‐specific transgene integration, suitable for a variety of biotechnological applications. Biotechnol. The accumulation of beneficial mutations on competing genetic backgrounds in rapidly adapting populations has a striking impact on evolutionary dynamics. This effect, known as clonal interference, causes erratic fluctuations in the frequencies of observed mutations, randomizes the fixation times of successful mutations, and leaves distinct signatures on patterns of genetic variation. Here, we show how this form of “genetic draft” affects the forward-time dynamics of site frequencies in rapidly adapting asexual populations. We calculate the probability that mutations at individual sites shift in frequency over a characteristic timescale, extending Gillespie’s original model of draft to the case where many strongly selected beneficial mutations segregate simultaneously. We then derive the sojourn time of mutant alleles, the expected fixation time of successful mutants, and the site frequency spectrum of beneficial and neutral mutations. Finally, we show how this form of draft affects inferences in the McDonald–Kreitman test and how it relates to recent observations that some aspects of genetic diversity are described by the Bolthausen–Sznitman coalescent in the limit of very rapid adaptation.  相似文献

Random T–DNA integration into the plant host genome can be problematic for a variety of reasons, including potentially variable transgene expression as a result of different integration positions and multiple T–DNA copies, the risk of mutating the host genome and the difficulty of stacking well‐defined traits. Therefore, recombination systems have been proposed to integrate the T–DNA at a pre‐selected site in the host genome. Here, we demonstrate the capacity of the ?C31 integrase (INT) for efficient targeted T–DNA integration. Moreover, we show that the iterative site‐specific integration system (ISSI), which combines the activities of the CRE recombinase and INT, enables the targeting of genes to a pre‐selected site with the concomitant removal of the resident selectable marker. To begin, plants expressing both the CRE and INT recombinase and containing the target attP site were constructed. These plants were supertransformed with a T–DNA vector harboring the loxP site, the attB sites, a selectable marker and an expression cassette encoding a reporter protein. Three out of the 35 transformants obtained (9%) showed transgenerational site‐specific integration (SSI) of this T–DNA and removal of the resident selectable marker, as demonstrated by PCR, Southern blot and segregation analysis. In conclusion, our results show the applicability of the ISSI system for precise and targeted Agrobacterium‐mediated integration, allowing the serial integration of transgenic DNA sequences in plants.  相似文献

Summary

Populations of rare clonally spreading species are increasingly being shown to be composed of one or a few genotypes. Clones have potentially unlimited life but two factors, genetic erosion due to random events and increasing accumulation of genetic load in older genotypes, combine to expose them to sexual failure and loss of fertility. Non-breeding clonal populations are at risk of extinction because they lack the ability to adapt or escape from changing environments.

Twinflower (Linnaea borealis L.) has been lost from nearly 50 % of its pre-1970 sites. It is a clonal self-incompatible plant and sexual failure has been reported from Canada, Britain and Scandinavia. Sexual failure is due to high levels of within-clone pollination by flies. Seed set occurs naturally in Scottish populations which have some genetic diversity. A non-breeding population in Scotland has been restored to viability by experimental cross-pollination with pollen from a distant source. Conservation action for this species should therefore be directed to the prevention of loss of habitat and restoration of population viability by translocation of compatible mates.

Translocating plants into extant, but non-breeding, populations are still a matter of debate and are frequently opposed. However, experimental translocations must now be performed to establish whether intervention can secure the long-term survival of rare clonal plants without the constant need for re-introduction.  相似文献
